Validation of KASP assays associated with barley adaptation and productivity traits

Barley (Hordeum vulgare L.) is an important cereal crop traditionally used in animal feed, malting, and food production. In Kazakhstan, barley the second most cultivated grain. However, despite long history of cultivation traditional breeding methods prevail here. The introduction marker-assisted selection (MAS) process may improve adaptation productivity local cultivars, as well help development new ones. For that, validation 23 kompetitive allele-specific PCR (KASP) assays for traits developed using previous GWAS results was performed. collection 35 two-rowed promising lines grown at Kazakh research institute agriculture plant growing (KRIAPG, Almaty region, Kazakhstan) 2021 studied 5 adaptation-related (heading time, heading-maturity height, peduncle length, spike length) productivity-related (number kernels per spike, weight plant, thousand weight, yield m2) traits. same genotyped KASP assays. As a result, 21 KASPs demonstrated good level polymorphism (MAF > 0.10 I 0.36) collection. Six confirmed their associations with (P < 0.05); nine were associated other agronomic 0.05).
